The present invention provides a mutant of E. coli heat-labile toxin and method of manufacturing the same. The manufacturing method and products thereof are performed by conducting site-directed mutagenesis on E. coli heat-labile toxin genes, replacing signal peptides of an A subunit and a B subunit included in the E. coli heat-labile toxin with signal peptides of E. coli outer membrane protein, and modifying signail peptides of E. coli outer membrane protein. In addition, the present invention has advantages including applying that in adjuvants and vaccines, making the same mass production, reducing the costs, and the like.
